From the Japanese, the term "big brother" is also something a little girl might call an older male whom she's friendly with. Since Catherina takes the form of a creepy little girl, I'd imagine this is the kind of thing she'd say in a creepy voice to some hapless person who decided to check out her mansion... Right before she erased his memory.

I'm going to agree with Logress on this, I got the feeling she was using it more as a term for any unsuspecting guy who wandered in, rather than a brother. (Especially since I have the feeling she's actually REALLY old, and any real brother she would have had wouldn't have been so gifted.)
